چکیده
Abstract Studies of immune responses elicited by bovine viral diarrhea virus (BVDV) vaccines have primarily focused on the characterization neutralizing B cell and CD4+ T epitopes. Despite availability commercial for decades, BVDV prevalence in cattle has remained largely unaffected. There is limited knowledge regarding role BVDV-specific CD8+ cells protection, indirect evidence suggests that they play a crucial during infection. In this study, presence are highly cross-reactive was demonstrated. Most importantly, novel potent IFN-γ–inducing epitopes were identified from different regions polyprotein. Eight following structural Ags: Erns, E1, E2 glycoproteins. addition, nonstructural Ags Npro, NS2-3, NS4A-B, NS5A-B, 20 identified. The majority these found to be conserved among more than 200 strains BVDV-1 -2 genotypes. These also validated as because induced high recall IFN-γ+CD8+ ex vivo purified isolated BVDV-1– -2–immunized cattle. Altogether, 28 MHC class I–binding key can elicit broadly reactive against diverse strains. data presented study will lay groundwork development contemporary cell–based vaccine capable addressing heterogeneity effectively current vaccines.

Persistent bovine viral diarrhea virus (BVDV) infection in cattle herds
Bovine viral diarrhea virus (BVDV) is a significant pathogen associated with gastrointestinal, respiratory, and reproductive diseases of cattle worldwide. It causes continuous economic losses to the cattle industry primarily due to decreased reproductive performance. The ability of virus to cross the placenta during early pregnancy can result in the birth of persistently infected (PI) calves. P...
متن کاملDetection of Bovine Viral Diarrhea Virus in Bovine Semen Using Nested-PCR
A rapid and sensitive reverse transcription polymerase chain reaction (RT-PCR) and nested-PCR were used to detect bovine viral diarrhea virus 1 (BVDV-1) in bull semen. Selected primers could amplify a part of the 5´UTR of the BVDV genome. A 294 bp DNA fragment was amplified and specificity of the results was confirmed by direct sequencing of the PCR product. Prior to RNA extraction, the seminal...
متن کاملWhole-Inactivated Influenza Virus Is a Potent Adjuvant for Influenza Peptides Containing CD8+ T Cell Epitopes
Influenza peptide antigens coding for conserved T cell epitopes have the capacity to induce cross-protective influenza-specific immunity. Short peptide antigens used as a vaccine, however, often show poor immunogenicity. In this study, we demonstrate that whole-inactivated influenza virus (WIV) acts as an adjuvant for influenza peptide antigens, as shown by the induction of peptide-specific CD8...
متن کاملThe effect of bovine viral diarrhea virus on bovine monocyte phenotype
Bovine viral diarrhea virus (BVDV) is an economically important pathogen of the livestock industry worldwide. BVDV is classified into cytopathic (cp) and noncytopathic (ncp), depending on its effects on cultured cells. BVDV is known to alter the host’s immune response. Of this, major histocompatibility complex (MHC) class II molecules play a central role in the development and function of the i...
متن کاملAntigenic variation among bovine viral diarrhea virus (BVDV) strains and the role of different cell fixation methods in immunoassays.
Antigenic variation among 13 Quebec isolates of bovine viral diarrhea virus (BVDV), 4 reference strains and 2 American isolates were studied by peroxidase-linked antibody assay (PLA assay) and neutralization test (NT). The Quebec strains consisted of 3 isolates before 1993 and 10 isolates from 1993. In the PLA assay, we compared 2 different fixatives, acetone and formalin. Acetone-fixation allo...
